横紋筋肉腫 / Rhabdomyosarcoma
横紋筋肉腫は、小児で最も高頻度に発生する軟部悪性腫瘍です (Sun 2015)。その一部は、筋芽細胞が起源と考えられています (Rubin 2011)。横紋筋肉腫の増殖や転移のメカニズムを明らかにし、その治療法や抗腫瘍剤を開発するには、筋芽細胞の研究成果が役に立ちます。私たちは、筋芽細胞の分化を促進する分子が、横紋筋肉腫の増殖を抑制するのではないかと考えて実験を進めています。
Rhabdomyosarcoma (RMS) is the most common sporadic soft-tissue sarcoma in children (Sun 2015). Some RMS are assumed to originate from myoblasts (Rubin 2011). The studies of myoblasts are useful to clarify the mechanisms of growth and metastasis of RMS, and finally to develop the novel therapeutic drugs for RMS. We are investigating whether the molecules which promote myoblast differentiation can prevent the growth of RMS.

横紋筋肉腫細胞の3次元培養 / 3D culture of RMS cells
抗腫瘍分子の作用を正確に評価するには、生体内に近い条件で腫瘍細胞を培養する必要があります (He 2017)。私たちは、横紋筋肉腫細胞を3次元培養し、増殖する腫瘍塊を形成することに成功しました (Shinji 2020)。この培養系を用いて、横紋筋肉腫の増殖メカニズムの解明や治療法の開発に貢献したいと考えています。
For precise evaluation of the effects of anti-tumor molecules, tumor cells need to be cultured under the conditions close to in vivo (He 2017). We successfully generated the tumorspheres of RMS cells which continuously grow in 3D-culture system (Shinji 2020). This system will be useful to study the mechanism of RMS growth and to develop the therapeutic strategy for RMS.
