Alkaline phosphatase treatment for DNA
Material
- Antarctic phosphatase (NEB M0289, -20C)
- x10 antarctic phosphatase reaction buf (NEB B0289, -20C)
Protocol
- Add 1/10 vol of x10 antarctic phophatase reaction buf to DNA (1-5 ug) sol.
- Add 1 ul of antarctic phosphatase.
- Tap to mix.
- Incubate at 37C for 15 min (5' extension or blunt) or 60 min (3' extension).
- Inactivate at 70C for 5 min.
